Having Duke be the ultra-nerdy guy is a nice fit, as well as making Maryland the comic book guy (really corny wannabes) also make pretty good sense IMO.

Miami should be Snake, but otherwise, well done.
Prof. Frink is way too cool to be a Duke guy, glavin, but the reference fits, so I'll go with it.
I'd have had the squeaky-voiced teen as Duke, but then I wouldn't know what to do with Clemson. Very good job all around.

Georgetown, of course, would be Krusty the Clown, the greatest entertainer in the world (well, except for Johnny Carson).
I don't know about the rest of the league, but in honor of their coach, I think Syracuse should be represented by Hans Moleman.
"Hello, this is Moleman in the morning. Good Moleman to you. Today part four of our series of the agonizing pain in which I live every daaaay."
Villanova should be represented by their guest lecturer, Barney Gumble.

West Virginia would be Cletus, of course.
Rutgers is Ralph Wiggum.

Louisville would be represented by Rick Pitino as the Monorail salesman.

You mean Lyle Lanley?
?: "I hear those things are awfully loud?"
LL: "It glides as softly as a cloud."
Apu: "Is there a chance the track could bend?"
LL: "Not on my life, my Hindu friend."
Barney: "What about us drunken slobs?"
LL: "You'll be given cushy jobs!"
Wiggum: "The ring came off my pudding can."
LL: "Use my pen knife, my good man."
Grampa: "Were you sent here by the devil?"
LL: "No good sir, I'm on the level."
Of course, there's the line that wins the crowd over in the first place: "You know the town with money is kinda like a mule with a spinning wheel: no one knows how he got it, and damned if he knows how to use it."

Cincinatti should be the criminal/ex-con character (snake?) due to all their trouble with the law.
It's harder to do the big east, as many of our schools such as Marquette, Depaul, Providence, SHU, St. Johns, and USF don't have very distinct fanbases.

Damn you people!
How the hell am I supposed to work with a conversation like this going on here? I'll be hitting up everyone on this board for a job when I get fired, count on it!
Anyway...
Cincinnati = the sleazy Vegas wives, or Cowboy Bob from Bob's RV Round-Up, or Dr. Nick Riviera; really just about any shady character
South Florida = Jasper (or Mrs. Glick)
Providence = Reverend Lovejoy, or 'The Leader, the head of the Movementarians.
UConn (aka ESPNU, aka U-Con) = Malloy, the Springfield Cat Burglar
Seton Hall = Jimbo or Kearney (not Nelson Muntz - he's too relevant)
SJU = Fat Tony, of course. Or Frankie the Squealer.
Marquette is a tough one, I don't know much about them. Let's go with Sir Oinksalot, the pig mascot.
Notre Dame = Ned Flanders as Satan from the Halloween episode?
Pitt = one of the gay steel workers from "The Anvil"
Anyone I missed who hasn't been covered yet?

Here's a tenative list so far... I'm working on the picture, could finish it today or tomorrow:
'Cuse - Moleman SJU - Fat Tony Prov - Rev. Lovejoy Cincy - Snake SFla - Jaspers Seton Hall - Kearney (can't be Jimbo, Jimbo actually gets some hot chicks, remember?) Pitt - Otto WVU - Cletus Rutgers - Ralph Wiggum
We've got some decisions to narrow down:
GU - God, Sideshow Bob or Krusty or Homer (hey, he runs the show) ND - Flanders or Martin Prince L'Ville - Lyle Lanley, Cowboy Bob, Jacques (any of those slickster, salseman types)
Which leaves us with four question marks:
UConn - that slick, crook of a New Englander, Diamond Joe Quimby? 'Nova - maybe someone nice and cheesey like Disco Stu? Kent Brockman and his over inflated sense of self-worth? DePaul - maybe Barney in homage to Chicago's boozing and eating habits? Marquette - maybe Wolfcastle in homage to Wisconsin's German heritage?
The picture will be up eventually... help me finish the list for now.

St. Johns - Nelson Muntz
Rutgers - The teenager with the squeaky voice

West Virginia - Cleetus
Notre Dame - Flanders
Pitt - Jimbo Jones
Nova - Frank Grimes

Kent Brockman for Nova is hysterical. They are definitely the "Sam the Eagle" of the Big East.

I think Grandpa Simpson would be good for Rutgers as well...no one actually listens to them but they love to talk.

Grandpa Simpson's classic tale:
"We can’t bust heads like we used to, but we have our ways. One trick is to tell ‘em stories that don’t go anywhere. Like the time I caught the ferry over to Shelbyville. I needed a new heel for my shoe, so, I decided to go to Morganville, which is what they called Shelbyville in those days. So I tied an onion to my belt, which was the style at the time. Now, to take the ferry cost a nickel, and in those days, nickels had pictures of bumblebees on ‘em. "Give me five bees for a quarter," you’d say. Now where were we? Oh yeah - the important thing was that I had an onion on my belt, which was the style at the time..."
